    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human SH2D2A/TSAd protein.

    Background

    The T cell-specific adapter protein (TSAd), encoded by the SH2D2A gene, is an adapter molecule containing an SRC homology 2 (SH2) domain and a proline-rich region (PRR) in addition to several tyrosine phosphorylation sites (1). The protein is predominantly expressed in T cells, NK cells, and endothelial cells (2,3). TSAd is required for T cell polarity establishment, interaction with antigen-presenting cells (4), and maintenance of T regulatory cell (Treg) function (5). The protein modulates T cell receptor (TCR) signaling in activated T cells via interaction with the tyrosine kinases Lck (6) and Itk (7). In endothelial cells, TSAd is required for VEGF-induced angiogenic sprouting through its binding to phosphorylated VEGF Receptor 2 (VEGFR2) at Tyr951 and coupling of VEGFR2 signaling to c-Src activation (8,9).
