Background
Secernin-1, encoded by the gene SCRN1, is a 50 kDa cytosolic protein implicated in a variety of biological functions. Originally discovered as a protein that regulates mast cell exocytosis, secernin-1 has been suggested to play roles in axon growth, endoplasmic reticulum (ER) remodeling, and presynaptic vesicle recycling (1-3). In Alzheimer’s disease (AD), secernin-1 was found to be abundant within amyloid plaques of dystrophic neurites and in neurofibrillary tangles (NFTs). Co-immunoprecipitation of secernin-1 and phosphorylated tau from AD patient brain samples further suggests a direct interaction between these proteins (4,5). Secernin-1 levels were also found to be increased in the cerebrospinal fluid (CSF) of AD patients, correlating with total and phospho-tau levels, suggesting its potential as a diagnostic and prognostic biofluid biomarker (6). Beyond AD, SCRN1 has been identified as a tumor-associated gene, with expression levels implicated in the development and progression of various cancers (7-9).
