    Background

    Ras-like without CAAX 1 (RIT1) is a member of the Ras superfamily of small GTPases (1,2). RIT1 promotes cell proliferation, survival, and stress responses via the activation of downstream signaling pathways p38 MAPK, ERK, Akt, and PAK1 (3-6). During mitosis, RIT1 interacts with and sequesters MAD2 from the mitotic checkpoint complex, leading to chromosome segregation errors and aneuploidy (7). The abundance of RIT1 is downregulated by ubiquitination and proteasomal degradation via the E3 ubiquitin ligase complex CRL3/LZTR1 (8). On the other hand, the RIT1 protein level is maintained by the deubiquitinase USP9X (9). RIT1 gene mutations result in RIT1 gain-of-function and have been identified in patients with Noonan syndrome (10,11). RIT1 gene amplifications and activation mutations are associated with and play an oncogenic driver role in lung adenocarcinoma (12), myeloid malignancies (13), hepatocellular carcinoma (14), and endometrial carcinoma (15).
