    Specificity / Sensitivity

    Phospho-Rab10 (Thr73) (F3X5B) Rabbit Monoclonal Antibody recognizes endogenous levels of Rab10 protein only when phosphorylated at Thr73. This antibody does not cross-react with other Rab proteins. This antibody detects a 300 kDa protein of unknown identity in some cell lines.

    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Thr73 of human Rab10 protein.

    Background

    Rab10 is a member of the Ras superfamily of small Rab GTPases (1) that interacts with Mss4, myosin V (Va, Vb and Vc) and GDI as it helps mediate sorting among cellular endosomes (2-4). Mutation analysis and GFP-fusion protein expression of Rab10 in MDCK cells determined that Rab10 plays a regulatory role in membrane protein transport between early endosomes and basolateral compartments (5,6). Rab10 associates with the GLUT4 complex as a target for AS160 and is required for insulin-stimulated GLUT4 translocation in adipocytes (7,8).

    Leucine-rich repeat kinase 2 (LRRK2) phosphorylates Rab10 at Thr73. LRRK2 hyperactivity, driven by pathogenic mutations or by the vacuolar protein sorting-associated protein 35 (VPS35) D620N Parkinson's disease (PD) associated mutation increases Rab10 phosphorylation at Thr73. Conversely, LRRK2 inhibition decreases Rab10 phosphorylation at this site, making it a key readout for LRRK2 activity (9-11).
