    Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    Phospho-PFK-2 (Ser466) Antibody detects endogenous Ser466 phosphorylated cardiac PFK-2, as well as the kidney isoform. This antibody may also reacts with the inducible PFK-2, as well as the brain, placental isoforms.

    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic phosphopeptide corresponding to residues surrounding serine 466 of human PFK-2 (cardiac isoform). Antibodies are purified by protein A and peptide affinity chromatography.

    Background

    PFK-2 (6-phosphofructo-2-kinase/fructose-2,6-bisphosphate 2-phosphatase) is a bifunctional enzyme catalyzing the synthesis and the degradation of fructose 2,6-bisphosphate, which is the potent stimulator of 6-phosphofructo-1-kinase and glycolysis. Several tissue specific isoforms have been reported, which have conserved catalytic core flanked by variable N- and C-terminal regulatory domains (1). It has been reported that Ser466 of the cardiac PFK-2, and Ser461 of the placental PFK-2 (also known as inducible PFK-2, iPFK-2) are phosphorylated by AMPK as well as by other kinases, which stimulates the activity of PFK-2 and glycolysis during ischemia and hypoxia (2-4). iPFK-2 process a typical oncogene-like regulatory element in its mRNA, which contributes to the high level expression of iPFK-2 and enhanced glycolysis in cancer cells (5).
