    Background

    Nuclear protein of the ataxia-telangiectasia locus (NPAT) is a transcriptional coactivator that is crucial for cell cycle progression (1,2). NPAT expression is regulated by the E2F transcription factor and its protein levels peak at the G1/S boundary (2,3). NPAT regulates the expression of all five histone gene subtypes (H1, H2A, H2B, H3, and H4) during the transition from G1 to S phase of the cell cycle (4,5). NPAT serves as a scaffolding protein for the formation of histone locus bodies (HLBs), which are unique biomolecular condensates functioning as localized hubs for histone mRNA synthesis and processing (6). Phosphorylation of NPAT by cyclin-dependent kinase 2 (CDK2) is required for histone mRNA transcription, but it is not necessary for HLB formation (7).  Mutations in the NPAT gene have been linked to various cancers, including colorectal cancer and Hodgkin lymphoma (8,9).
