    Background

    Caspase recruitment domain-containing protein 8 (CARD8) is a widely expressed protein that helps to sense and regulate inflammation (1). CARD8 is autoproteolytically processed between Phe296 and Ser297 into N- and C-terminal fragments that non-covalently associate to form an inactive complex with DPP9 under homeostatic conditions (2-4). In response to proteotoxic stress, however, the C-terminal fragments are released and assemble into inflammasomes, leading to caspase-1 recruitment and pyroptosis (1,5). HIV-1 protease is a major known activator of CARD8, cleaving CARD8 upstream of its autoproteolytic cleavage site to induce pyroptosis (6). In addition to its pro-inflammatory roles, CARD8 can also inhibit inflammation by blocking NLRP3 and NF-κB activity (7,8). Unlike its highly conserved homolog, NLRP1, CARD8 does not have rodent orthologs (9).
