    Background

    Acyl-CoA-binding domain-containing protein 3 (ACBD3) is a ubiquitously expressed Golgi resident protein that is involved in lipid metabolism, membrane trafficking, and Golgi structure maintenance (1). ACBD3 is localized to the Golgi through interaction with giantin/GOLGB1 and functions as an adaptor protein, recruiting additional proteins, such as golgin-160/GOLGA3, to regulate apoptosis and Dexras1 to maintain iron homeostasis (2-4). ACBD3 also recruits phosphatidylinositol 4-kinase beta (PI4KB) to the Golgi, which is critical for regulating vesicle formation and lipid transport (5,6). Several enteroviruses utilize ACBD3 for efficient replication via interaction with PI4KB and the viral 3A protein, and depletion of ACBD3 has been shown to impair viral RNA synthesis (7,8). ACBD3 is also associated with Huntington’s disease (HD), in which elevated ACBD3 protein levels are observed in the striatum of HD patients, and its knockdown eliminates HD-associated neurotoxicity in cell models (9).
