    Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    Phospho-(Thr) PDK1 Substrate Antibody detects phosphorylated threonine in the T*FCGT motif. It does not cross-react with nonphosphorylated threonine residues, phospho-serine residues in the same motif or phospho-threonine in other motifs. Phospho-(Thr) PDK1 Substrate Antibody detects endogenous levels of many phosphorylated proteins including phospho-PKC isoforms and phospho-Akt (Thr308).     Source / Purification

    Polyclonal antibodies are produced by immunizing animals with peptides containing a phospho-threonine in T*FCGT motif. Antibodies are purified by protein A and peptide affinity chromatography.

    Background

    The AGC subfamily of protein kinases, including Akt, p70 S6 kinase, protein kinase C isoforms, p90RSK and SGK, has two phosphorylation sites (1). One is located in the T-loop of the kinase domain (1,2). The other phosphorylation site is located in the carboxy-terminal hydrophobic motif (1). Phosphorylation of these sites is necessary for kinase activity (1-3). Although 3-phosphoinositide-dependent kinase 1 (PDK1) has been shown to phosphorylate the T-loop of many members of the AGC subfamily of protein kinases in a sequence motif comprising T*FCGT, where the first threonine residue becomes phosphorylated in response to cell stimulation (1), many of its substrates remain unidentified. In order to identify potential PDK1 substrates, CST has developed antibodies that recognize phosphorylatedthreonine in T*FCGT motifs. As shown by ELISA, Phospho-(Thr) PDK1 Substrate Antibody is specific to PDK1 substrates containing phospho-threonine.
