    Protocol

    Specificity / Sensitivity

HuD/ELAVL4 (F4F3X) Rabbit mAb recognizes endogenous levels of total HuD/ELAVL4 protein. Species reactivity for immunofluorescence is mouse only.

    Species Reactivity:

    Mouse, Rat

The antigen sequence used to produce this antibody shares 100% sequence homology with the species listed here, but reactivity has not been tested or confirmed to work by CST. Use of this product with these species is not covered under our Product Performance Guarantee.

Species predicted to react based on 100% sequence homology:

    Human

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro270 of human HuD/ELAVL4 protein.

    Background

The embryonic lethal, abnormal vision, and Drosophila-like (ELAVL) family of RNA-binding proteins has four members that undertake essential functions in brain development. ELAVL1 is ubiquitously expressed and predominantly involved in stabilizing mRNA targets. HuR/ELAVL1 is crucial for cellular processes such as cell proliferation, differentiation, and stress response (1). HuB/ELAVL2, HuC/ELAVL3, and HuD/ELAVL4 have historically been more specifically associated with neurodevelopment, with expression enriched in the central and peripheral nervous system, and are therefore referred to as neuronal ELAVLs (nELAVLs) (1). HuD/ELAVL4 is predominantly expressed in neurons and, at lower levels, in the pancreas and testis (2). It is associated with type 2 diabetes mellitus and various neurodegenerative diseases (3). The level of HuD/ELAVL4 was shown to be decreased in the hippocampus of Alzheimer’s disease (AD) patients, and it regulates the translation and splicing of multiple AD candidate genes (3,4). From genetic studies, HuD/ELAVL4 was also identified as a susceptibility gene for Parkinson’s disease (PD) (5,6).
