    Product Usage Information

    This product is the carrier free version of product #44783. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

    This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

    BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity.

    Formulation

    Supplied in 1X PBS (10 mM Na2HPO4, 3 mM KCl, 2 mM KH2PO4, and 140 mM NaCl (pH 7.8)). BSA and Azide Free.

    Storage

    Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

    Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the serine protease and PDZ domains of human HTRA1/PRSS11 protein.

    Background

    High-temperature requirement A serine peptidase 1 (HTRA1, PRSS11) is a trypsin-like serine protease involved in neuronal maturation, protein degradation, apoptosis, and the response to cellular stress. HTRA family members contain N-terminal IGFBP and Kazal-like domains, a serine protease domain, and a C-terminal PDZ domain (1). HTRA1 can be secreted and autocatalyzed (2) or retained in the cell and processed to shorter cytoplasmic- and nuclear-localized forms (3). Ser328Ala mutants lack both proteolytic and autocatalytic activity (2). HTRA1 regulates the extracellular matrix (ECM) by degrading aggrecan, collagen, decorin, fibromodulin, and fibronectin (4,5), while also breaking down signaling molecules such as TGF-β family members (6,7) and FGF8 (8). In the brain, HTRA1 degrades β-Amyloid and aggregated fibrillar tau (9,10). Elevated HTRA1 can contribute to osteoarthritis (5,11) and age-related macular degeneration (12). HTRA1 loss induces EMT and breast cancer cell invasion (13).
