Background
Four and a half LIM-only protein 2 (FHL2) is a member of the LIM domain protein family that interacts with a large number of proteins, including receptors, signal transducers, and transcription factors (1). LIM proteins have one or more LIM domains composed of a conserved cysteine-rich module that mediates protein interactions (2). FHL2 is involved in regulating various cellular processes, such as survival, proliferation, migration, and signal transduction in muscle and cardiac tissue (1-3). In immune cells, FHL2 has been shown to play a role in B cell differentiation and maturation in secondary lymphoid organs, and its deletion alters cytokine release and migration of dendritic cells and macrophages (4,5). FHL2 binds FOXO1, resulting in decreased expression of genes targeted by FOXO (6). FHL2 suppresses tumor growth by interacting directly with HIF-1α and repressing its transcriptional activity (7). When upregulated in tumors, FHL2 can also act as an oncoprotein interacting with multiple transcription factors to suppress apoptosis and promote tumor progression (8). For these reasons, FHL2 is being investigated as a tumor therapeutic target.
