Background
DNA polymerase β (POLB) functions in the base excision repair (BER) DNA repair pathway, repairing single-strand DNA breaks (SSBs) arising from oxidative damage, deamination, alkylation, or abasic sites (1). In mitochondria, oxidative stress causes recruitment of DNA polymerase β to repair damaged mitochondrial DNA (mtDNA) (2).
DNA polymerase β expression levels and mutations are associated with human cancer (3-6). Inhibition or downregulation of DNA polymerase β is synthetic lethal with defective mismatch repair (MMR) in models of acute lymphoblastic leukemia (ALL) (7) and with BRCA1 deficiency in models of breast cancer (8).
