Specificity / Sensitivity

FHIT Antibody detects endogenous levels of total FHIT protein.

Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to amino acids near the amino terminus of human FHIT. Antibodies are purified by protein A and peptide affinity chromatography.

Background

The fragile histidine triad (FHIT) tumor suppressor gene is frequently altered in human tumors (1). Adenovirus-mediated FHIT gene transfer has been shown to arrest tumor cell growth and induce apoptosis (2,3), and FHIT-/- knockout mice have a higher incidence of spontaneous tumor formation (4,5). FHIT physically associates with ubiquitin-conjugating enzyme 9 (6), and Src has been reported to phosphorylate Tyr114 of FHIT in vitro and in vivo (7). FHIT is a diadenosine P1,P3-triphosphate hydrolase (8), but a His96Asn mutation, which reduces FHIT enzymatic activity by three-fold, does not reduce FHIT tumor suppressor activity (8,9).
