Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro57 of human SIGMAR2/TMEM97 protein.

Background

Sigma intracellular receptor 2 (SIGMAR2 or TMEM97) is an endoplasmic reticulum (ER)-resident membrane chaperone protein that regulates the sterol transporter NPC1 (1). Recently, SIGMAR2/TMEM97 was identified as the opioid 2 receptor. Opioid receptors are a large super family of 7 transmembrane-spanning G protein-coupled receptors (GPCRs) that are activated by both endogenous opioid peptides and by exogenously administered opiate compounds (2). Opioid receptors are of pharmacological interest not only as analgesics but to treat neurological disorders including psychiatric and neurodegenerative diseases. Opioid receptors are classified pharmacologically, including σ1 and σ2 type receptors. Although SIGMAR1 was identified in 1996 as the σ1 receptor, identification of the σ2 type receptor remained elusive until 2017 when TMEM97 was identified as the σ2 type receptor (3,4). In addition to acting as an opioid receptor, SIGMAR2/TMEM97 may also act within a complex to regulate cellular update of Aβ, the pathological hallmark of Alzheimer's disease (5,6).
