Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

RUFY3 Antibody recognizes endogenous levels of total RUFY3 protein. RUFY3 Antibody detects multiple RUFY3 isoforms reported to be expressed by alternative splicing, including human RUFY3 isoform 1 and 2.

Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding His58 of human RUFY3 protein. Antibodies are purified by peptide affinity chromatography.

Background

RUN and FYVE domain-containing protein 3 (RUFY3) is an actin-binding protein that is enriched in the brain. RUFY3 was identified as a protein that is upregulated during neuronal polarization, which is important for proper neuron functioning (1). RUFY3 likely functions in regulating neuronal polarization by interacting with the neuronal cytoskeleton via actin filament-binding proteins, such as Fascin (2). RUFY3 undergoes various forms of posttranslational modifications, including CASP3-dependent cleavage and phosphorylation, which may modulate selective synaptic and axonal degeneration during brain development and neurodegenerative disease (3).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
