Background

Partner and localizer of BRCA2 (PALB2), also known as Fanconi Anemia Complementation Group N (FANCN) protein, is an essential binding partner for BRCA1 and BRCA2 proteins. PALB2 acts as a molecular scaffold for the BRCA1/BRCA2/RAD51 DNA repair complex, and functions to help recruit this complex to sites of DNA damage and stimulate RAD51 DNA strand-invasion activity required for D-loop formation (1). PALB2 is a tumor suppressor protein that is required for homologous repair of double-stranded DNA breaks. Mutations in PALB2 (mostly mono-allelic truncations) lead to increased susceptibility to breast, prostate, and pancreatic cancers (2-4). Homozygous mutation of PALB2 leads to FANCN, a disorder affecting all bone marrow elements, resulting in anemia, leukopenia, and thrombopenia (5).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
