Background

Olaparib (AZD2281) is a potent and selective PARP-inhibitor, specifically targeting PARP1 and PARP2 (IC50 = 5 nM and 1 nM, respectively) (1). This small molecule has synergistic effects on breast cancer cell lines when combined with other treatments, independent of BRCA1 status (2). Non-small cell lung cancer (NSCLC) cancer cells that are homologous recombination (HR)-deficient are hypersensitive to Olaparib (AZD2281) treatment, resulting in reduced cell viability (3).

Storage

Store lyophilized at -20ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 1 month to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

Directions for Use

Olaparib (AZD2281) is supplied as a lyophilized powder. For a 10 mM stock, reconstitute 5 mg of powder in 1.15 ml of DMSO. Working concentrations and length of treatment can vary depending on the desired effect.
