Background

Zinc finger proteins (ZNFs) are one of the largest groups of transcriptional regulatory proteins and consequently are involved in a wide range of cellular processes (1,2). ZNFs have been implicated in transcriptional regulation, ubiquitin-mediated protein degradation, signal transduction, actin targeting, DNA repair, as well as cell proliferation, migration, and apoptosis (2). ZNFs have been shown to interact with DNA, RNA, poly-ADP-ribose (PAR), and other proteins (1,2). ZNF148, also known as ZBP89, was first discovered and characterized as a C2H2 family transcription factor (1) and has been primarily studied for its role in cancer cell biology. Several tumor types, such as breast, liver, skin, brain, and gastric cancers, all exhibit increased ZNF148 expression versus normal tissue, suggesting an oncogenic function (3-9). Conversely, ZNF148 has also been shown to act as a tumor suppressor in colorectal cancers by repressing cell proliferation and inducing apoptosis (4,5). The likely reason for these dual roles for ZNF148 is that it has two functionally distinct alternative splicing isoforms: one encoding the full-length protein, and one that lacks the N-terminal domain (1,2). These two alternatively spliced isoforms have been shown to act antagonistically to each other (2). This dual regulatory role of ZNF148, along with its interaction with multiple proteins that facilitate critical aspects of tumor biology, make it a key protein of interest for cancer biologists.

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
